    Inorganic carbon uptake

    Abstract

    Protocol as descibed in version 2 of the AeN protocols on p 108 (Primary production). The aim of my work on the first AeN cruise was to estimate the uptake rate of CO2 via different mechanisms and in different habitats in the Arctic. The studied habitats included water samples from the CTD rosette, sediment samples from the box corer, sea ice samples from the lowermost 1-2 cm of the ice cores, brine fluid from sack holes, melt pond water and algae aggregates. CO2 uptake was measured as photosynthetic uptake by microalgae and cyanobacteria, as nitrification by nitrifying bacteria and archaea, and as anapleurotic reactions (CO2 uptake into the TCA cycle) of heterotrophic microorganisms.

    Ice meiofauna and protists

    Abstract

    Ice cores were taken with a 9 cm inner diameter Kovacs ice corer. Ice cores were melted at 4 deg C with added filtered seawater (100 ml per 1 cm core section). Total volume of the melted sample was measured, the sample gently mixed, and 100 ml filled in brown glass bottles and preserved with 1 ml GAA Lugol for protist analysis. The remaining volume was sieved over 64 um (note: 20 um sieve requested in protocol was not available), rinsed into vials with a squeeze bottle and preserved in ~2-4% Zoofix for meiofauna analysis. Protocol 15.5.7 from Sampling protocol version 1 was used, except that sections 0-1 and 1-3 cm were combined into one since 0-1 cm was impossible to saw in the melting conditions we experienced.

    Abstract

    This datasheet includes all samples obtained through box core sampling during Arven etter Nansen Joint Cruise 1-2, 6-23 August, 2018. Samples were collected for RF1 objectives (includes sediment grain size, porosity, trace elements) and RF3 objectives (pigments, stable isotopes, total organic carbon, meiofauna/foraminifera communities, juvenile epifauna communities, microbial communities and biomass production, phytoplankton spores).
